Product Description

Ultra-DNase I (RNase free) is a 29 kDa monomer. It is in routine use for removal of DNA from RNA and its related products. DNase I is a potent endonuclease which cleaves ssDNA or dsDNA nonspecifically to give oligonucleotides with 5’-PO4 and 3’-OH
groups.
Source: An E. coli strain that carries the cloned gene of bovine DNase I.

  • Activity: Potent endonuclease that nonspecifically cleaves ssDNA or dsDNA to oligonucleotides with 5′-PO₄ and 3′-OH ends

  • Unit definition: One unit completely degrades 1 µg plasmid DNA at 37 °C for 10 min in a 50 µL reaction

  • Heat inactivation: 75 °C for 10 min

  • Inhibition: Inhibited by metal chelators, transition metals (e.g., Zn), SDS (>0.1%), reducing agents (DTT, β-mercaptoethanol), and high ionic strength (>50–100 mM)

  • Purity: ≥95–98% purity confirmed by 10% reducing SDS-PAGE

  • RNase activity: >90% RNA remains intact after incubation, confirming RNase-free nature

  • Storage temperature: Store at –20 °C

  • Delivery form: Supplied in storage buffer containing 10 mM Tris-HCl (pH 7.5), 2 mM CaCl₂, and 50% glycerol
