HotStart Taq DNA Polymerase is a chemically modified thermostable enzyme designed for high-specificity DNA amplification. The enzyme originates from the Taq DNA Polymerase gene of Thermus aquaticus, which is expressed in E. coli. With a molecular weight of 94 kDa, it retains the characteristic 5' → 3' polymerase activity essential for PCR applications.

The HotStart modification ensures that the enzyme remains inactive at room temperature, thereby preventing non-specific amplification and primer-dimer formation during reaction setup. Upon heating to 95 °C during the initial denaturation step the enzyme is fully reactivated, enabling precise and efficient DNA synthesis in subsequent PCR cycles.

  • Purity: ≥98% purity confirmed by 10% reducing SDS-PAGE

  • Activity: Successful amplification of a 5 kb Lambda DNA fragment using 1.25 U enzyme in 25 PCR cycles.

  • RNase Activity: >90% of single-stranded RNA remains intact after 4 hours, indicating no RNase contamination.

  • DNase Activity: No detectable DNA degradation after 4 hours, confirming absence of DNase activity.

  • Storage Temperature: Store at –20 °C.
