It is routinely used in characterization of basic variants of monoclonal antibodies, as well as in production processes for molecules such as insulin. It is also used in mAb production processes for reducing the charge heterogeneity of the mAb material.
CPB products from Edna have exceptional activity, specificity, stability and value in terms of cost per unit. The product is recombinant and totally free of contaminants such as Carboxypeptidase A which are often present in non-recombinant products currently on the market. The enzyme is supplied in its active form containing a non-dialyzable Zinc ion to activate the metalloproteinase. It is also used in mAb production processes for reducing the charge heterogeneity of the mAb material.
CPB is typically isolated from bovine, porcine, rat or human pancreas. Edna CPB is produced by expressing the CPB gene from Rattus norvegicus in E. coli, allowing for expression of the rat enzyme without any of the contaminants associated with pancreatic isolation processes, such as Carboxypeptidase A and trypsin, which can significantly reduce the consistency and specificity of the enzyme product. The Edna process expresses the enzyme as an unfolded precursor, followed by in-vitro refolding and prosequence cleavage. These are followed by multiple chromatography steps to ensure that all the enzyme present in the vial is in its folded, maximally active form.
