Modified Trypsin, Frozen, Mass Spectrometry Grade

Isolated from Bos Taurus pancreas, TPCK-treated, Modified

Trypsin is a serine endopeptidase that selectively cleaves at the C-terminal of lysine and argnine residues. The product is highly purified with multiple chromatography steps and modified by reductive methylation to cap the ε-amino groups of lysine, and thus is extremely resistant to autolytic self-cleavage. The product is further treated with L-(tosylamido-2-phenyl) ethyl chloromethyl ketone (TPCK) to inactivate any chymotrypsin activity which may be present, without affecting trypsin activity.

Trypsin products from Edna have exceptional activity, specificity, stability and value in terms of cost per unit. Learn more in the Product Data Sheet and Brochure with product information, detailed usage protocols and sample results.

Product Information

Trypsin is a serine endopeptidase that selectively cleaves at the C-terminal of lysine and argnine residues. The product is highly purified with multiple chromatography steps and further treated with L-(tosylamido-2-phenyl) ethyl chloromethyl ketone (TPCK) to inactivate any chymotrypsin-like activity. The product is also modified by reductive methylation to cap the ε-amino groups of lysine and prevent autolytic self-cleavage reactions. Edna Trypsin products are resistant to mild denaturing conditions such as 0.1% SDS, 1M urea or 10% acetonitrile.

What's In The Box:

The product includes a vial of the frozen enzyme in 50 mM acetic acid and a vial of dilution buffer (50 mM acetic acid) which can be used to further dilute the enzyme, along with 2X Trypsin reaction buffer compatible with mass spectrometry systems.

Highlights:

  • Sourced from bovine pancreas and purified with multiple chromatography steps to >99% purity

  • High sequence coverage

  • Specificity improved by TPCK treatment to remove any chymotryptic activity (reduced to <10 ATEE units per mg)

  • Resistant to mild denaturing conditions such as 0.1% SDS, 1M urea or 10% acetonitrile

  • Supplied with 2X Reaction Buffer

  • Robust and reliable reactions

  • Use with EdnaPRO digestion improver (0.01–0.2% in reaction solution), a custom detergent for improved cleavage at hard-to-access sites

  • Suitable for digestion in both gel and solution

  • Compatible for co-digestion with other enzymes including PNGase F, Lysine-C, Glu-C, IdeS Protease, and more
