Product Information

T4 DNA Ligase catalyzes the formation of phosphodiester bonds between 5' and 3' termini in double stranded DNA using ATP as a cofactor. The enzyme can be used for ligation of both blunt and sticky ends as well as single-stranded DNA nick repair.

Edna T4 DNA Ligase products are recombinantly produced in E. coli and thus are free from contaminating nucleases, offering robust and reliable ligation activity.

What's In The Box:

The product includes one vial of recombinant enzyme at standard or high concentration, along with one vial of 10x ligation buffer.

50 mM Tris-HCl
10 mM MgCl2
1 mM ATP
10 mM DTT
pH 7.5 @ 25°C

Properties and Storage

Unit Definition: One unit is defined as the amount of enzyme required to give 50% ligation of HindIII fragments of DNA from Bacteriophage λ (5´ DNA termini concentration of 0.12 µM, 300- µg/ml) in a total reaction volume of 20 μl in 30 minutes at 16°C in 1x T4 DNA Ligase Reaction Buffer.

Heat Inactivation: >65 °C for 15 minutes
